Unveiling the clinical signs and pathology in red deer (Cervus elaphus) naturally infected with epizootic haemorrhagic disease virus serotype 8 https://gisaz.org/unveiling-the-clinical-signs-and-pathology-in-red-deer-cervus-elaphus-naturally-infected-with-epizootic-haemorrhagic-disease-virus-serotype-8 https://gisaz.org/unveiling-the-clinical-signs-and-pathology-in-red-deer-cervus-elaphus-naturally-infected-with-epizootic-haemorrhagic-disease-virus-serotype-8#respond Thu, 02 Jul 2026 19:16:20 +0000 https://gisaz.org/?p=1975 Abstract

Epizootic haemorrhagic disease (EHD) is a notifiable disease to the World Organisation for Animal Health caused by Orbivirus ruminantium (EHDV), closely related to bluetongue virus (BTV) and transmitted by Culicoides spp. In autumn 2022, EHDV serotype 8 (EHDV-8) was detected for the first time in Spain and Europe, affecting domestic and wild ruminants. Red deer (Cervus elaphus) were the most affected wild species, with reports of neurological signs, severe respiratory distress and mortality. Nevertheless, the pathogenesis of EHDV-8 in this species remains poorly understood. This study aimed to evaluate the clinical and pathological impact of EHD in red deer, characterising clinical signs, main lesions, tissue distribution and target cells of EHDV. Clinical data were collected through a nationwide online survey and field outbreak studies, and histopathological and immunohistochemical analyses were performed on 16 confirmed EHDV-8 cases. Frequent clinical signs included neurological signs, ptyalism, lameness and dyspnoea. During the outbreak, average morbidity and mortality in captive red deer were 6.9% and 4.9%, respectively, with a case fatality rate of 70.3%, higher in males and almost exclusively in adults. Gross examination revealed extensive vascular and epithelial lesions with generalised congestion, focal to multifocal haemorrhages and marked pulmonary oedema, confirmed histologically with severe cellular damage. The most affected organs were the brain, lungs, liver, kidneys and spleen, showing interstitial oedema, vasculitis, mononuclear infiltration with necrosis, severe lymphoid depletion and gliosis with satellitosis in the brain. The EHDV antigen was primarily detected in macrophages, reticular cells, endothelial cells and epithelial cells. This study provides the first description of EHDV’s clinical and pathological impact in red deer, confirming their high susceptibility to EHDV-8. https://doi.org/10.1186/s13567-026-01807-w

Leishmaniosis is a vector-borne zoonosis mainly caused by Leishmania infantum in Europe. Exposure to this protozoan has been widely documented in various domestic and wild species, which have been suggested as competent reservoirs of L. infantum in anthropogenic environments. Notably, wild lagomorphs, particularly the European wild rabbit (Oryctolagus cuniculus), have emerged as key natural reservoirs, linked to human leishmaniosis outbreaks in Spain. Despite their recognized role in the L. infantum epidemiology, significant knowledge gaps remain regarding the spatiotemporal dynamics of this pathogen. Consequently, this study aimed to evaluate the long-term circulation of L. infantum in wild rabbit populations in Mediterranean ecosystems of southern Spain. Thus, 1076 wild rabbits were analysed across five consecutive study periods: 2009–2012 (P1), 2012–2015 (P2), 2015–2018 (P3), 2018–2020 (P4), and 2020–2023 (P5), using the indirect immunofluorescence antibody test. Overall seroprevalence was 14.8% (159/1076; 95% CI: 12.7–16.9). Seropositivity proportions fluctuated but remained homogeneous over time (P = 0.10), ranging from 10.7% (P4) to 18.5% (P1). Spatiotemporal analysis identified two statistically significant clusters of L. infantum seropositivity within the study area. Although rabbit density was significantly associated with L. infantum seroprevalence, its overall contribution appears limited. This long-term surveillance reveals endemic, widespread, and spatially heterogeneous circulation of L. infantum in wild rabbit populations within the study region, underscoring their role as maintenance reservoirs in Iberian Mediterranean ecosystems. These findings provide key spatiotemporal insights into L. infantum dynamics, essential for designing and implementing effective surveillance and control strategies in high-risk areas. https://doi.org/10.1016/j.ijppaw.2026.101249

Beyond Genes: Metabolomic Evidence Indicates PotentialSpecies‐Level Differentiation in European Wild Rabbits https://gisaz.org/beyond-genes-metabolomic-evidence-indicates-potentialspecies%e2%80%90level-differentiation-in-european-wild-rabbits https://gisaz.org/beyond-genes-metabolomic-evidence-indicates-potentialspecies%e2%80%90level-differentiation-in-european-wild-rabbits#respond Thu, 28 May 2026 15:27:36 +0000 https://gisaz.org/?p=1981 ABSTRACT

The European rabbit (Oryctolagus cuniculus) plays a key ecological role in Mediterranean ecosystems, yet its populations aredeclining. Two subspecies, O. c. algirus and O. c. cuniculus, are present in the Iberian Peninsula and exhibit genetic, phenotypic, and ecological differences. The aim of this study was to investigate whether genetic differences between subspecies are reflectedin the plasma metabolome of the European rabbit and to identify potential biomarkers for subspecies classification. Com-prehensive untargeted metabolomics analyses (n = 49 wild individuals) were performed. Significant and consistent differencesin the metabolomic profiles were found between subspecies. Statistically significant differences were detected in nine metab-olites; three showed strong discriminatory potential, with Medicagenic acid, MG(P‐18:0/0:0/0:0), and Androsta‐1,4‐diene‐17‐carboxylicacid, 17‐[(ethoxycarbonyl)oxy]‐11‐hydroxy‐3‐oxo‐, fluoromethyl ester showing strong potential as biomarkers. These findings demonstrate functional divergence between the subspecies, likely related to diet and ecological adaptations. They provide new molecular evidence of metabolic differentiation between subspecies, which may be relevant for under-standing evolutionary divergence and could inform conservation strategies. 10.1002/jez.70097
